چکیده
زمینه و هدف: پیشرفت سرطان پستان با فعال‌سازی محور IGF-1 و افزایش استرس اکسیداتیو همراه است که می‌تواند تکثیر تومور را تسریع کند. تمرین مقاومتی (RT) و رزوراترول (RSV) احتمالاً از طریق تعدیل IGF-1 و بهبود وضعیت آنتی‌اکسیدانی، این اختلالات را کاهش می‌دهند. هدف این مطالعه بررسی اثر RT، RSV و RT+RSV بر IGF-1، مالون‌دی‌آلدئید (MDA) و ظرفیت آنتی‌اکسیدانی تام (TAC) در مدل موشی سرطان پستان بود.
روش تحقیق: : این مطالعه تجربی با طرح پس‌آزمون و گروه کنترل انجام شد. پنجاه موش ماده ویستار به‌صورت تصادفی در پنج گروه HC، CC، RT، RSV و RT+RSV تقسیم شدند. پروتکل تمرین مقاومتی به مدت 8 هفته، هفته‌ای 3 جلسه اجرا شد. رزوراترول نیز روزانه با دوز 25 میلی‌گرم به ازای هر کیلوگرم وزن بدن تجویز گردید. در پایان مداخله، بیان ژن IGF-1 با RT-qPCR و شاخص‌های MDA و TAC با روش‌های رنگ‌سنجی اندازه‌گیری شدند. داده‌ها با تحلیل واریانس دوعاملی بررسی شدند.
یافته‌ها: در گروه CC نسبت به گروه HC، افزایش IGF-1 و سطح MDA و کاهش TACمشاهده شد (001/0>p). هر دو مداخله RT و RSV به‌صورت مستقل موجب کاهش IGF-1 و MDA و افزایش TAC شدند (001/0>p)، و بیشترین بهبود در گروه ترکیبی RT+RSV مشاهده گردید. اثر اصلی تمرین و رزوراترول بر هر سه شاخص معنی‌دار بود (001/0>p)، در حالی‌که اثر تعاملی تمرین × رزوراترول تنها برای TAC معنی‌دار بود (001/0>p)
نتیجه‌گیری: ترکیب RT و مکمل RSV احتمالاً می‌تواند به‌عنوان یک راهبرد مکمل غیرتهاجمی در بهبود تعادل اکسیدان–آنتی‌اکسیدان و تعدیل محور IGF-1 در سرطان پستان مؤثر باشد.

Background and Aim: Breast cancer progression is associated with activation of the IGF-1 axis and increased oxidative stress, both of which may accelerate tumor proliferation. Resistance training (RT) and resveratrol (RSV) may attenuate these disturbances by modulating IGF-1 and improving antioxidant status. Therefore, the present study aimed to investigate the effects of RT, RSV, and combined RT+RSV on IGF-1, malondialdehyde (MDA), and total antioxidant capacity (TAC) in a murine model of breast cancer. Methods: This experimental study was conducted using a post-test design with a control group. Fifty female Wistar mice were randomly assigned to five groups: healthy control (HC), cancer control (CC), resistance training (RT), resveratrol supplementation (RSV), and combined resistance training plus resveratrol (RT+RSV). The resistance training protocol was performed for 8 weeks, three sessions per week. Resveratrol was administered daily at a dose of 25 mg/kg body weight. At the end of the intervention period, IGF-1 gene expression was assessed using RT-qPCR, while MDA and TAC levels were measured using colorimetric assays. Data were analyzed using two-way analysis of variance (ANOVA). Results: Compared with the HC group, the CC group exhibited significantly increased IGF-1 expression and MDA levels, along with a significant reduction in TAC (p < 0.001). Both RT and RSV interventions independently decreased IGF-1 expression and MDA levels while increasing TAC (p < 0.001), with the greatest improvement observed in the combined RT+RSV group. Significant main effects of resistance training and resveratrol were found for all three variables (p < 0.001), whereas the interaction effect of RT × RSV was significant only for TAC (p < 0.001). Conclusion: The combination of resistance training and resveratrol supplementation may serve as a complementary non-invasive strategy for improving oxidant–antioxidant balance and modulating the IGF-1 axis in breast cancer
