Background and Aim: Type 2 diabetes mellitus (T2DM) is associated with the induction of oxidative stress in the hippocampus, which is considered an important mechanism in diabetic neuropathy and cognitive impairment. The present study aimed to investigate the effect of eight weeks of aerobic training and cinnamon extract consumption on the levels of malondialdehyde, superoxide dismutase, and protein carbonyl in the hippocampus of a T2DM rat model. Materials AND Methods: Thirty-five male Wistar rats were divided into five groups (seven rats per group), including healthy control, diabetic control, diabetic + cinnamon extract, diabetic + aerobic exercise, and diabetic + aerobic exercise + cinnamon extract. Diabetes induction was performed by nicotinamide/streptozotocin injection. Interventions included eight weeks of moderate-intensity treadmill running (five sessions per week) and/or daily cinnamon extract (200 mg/kg) gavage. The levels of malondialdehyde (MDA), superoxide dismutase (SOD), and protein carbonyl (PC) in hippocampal tissue were measured by ELISA. In data analysis, one-way analysis of variance and Bonferroni post hoc test were used, observing all statistical assumptions (p ≤ 0.05). Results: The combined intervention (diabetic group + exercise + cinnamon extract) showed the greatest effect and significantly increased SOD enzyme activity compared to all diabetic groups. Also, MDA and PC levels in this group were significantly reduced compared to the diabetic control group (p=0.001). Each of the exercise training interventions and cinnamon extract consumption alone also improved these indicators compared to the diabetic control group (p=0.001), but the extent of this improvement was less than the effect observed in the combined protocol. onclusion: Eight weeks of aerobic training combined with cinnamon extract reduces hippocampal oxidative stress in T2DM, which may prevent the occurrence of central nervous system complications of diabetes, especially neuropathy. This strategy may be a promising complementary approach for the management of neurological complications of diabetes.
