Comparison of the effect of Resistance Training and Resveratrol Supplementation on IGF-1 Expression and Oxidative Stress Markers (MDA and TAC) in Mice with Breast Cancer

Abstract
Background and Aim: Breast cancer progression is associated with activation of the IGF-1 axis and increased oxidative stress, both of which may accelerate tumor proliferation. Resistance training (RT) and resveratrol (RSV) may attenuate these disturbances by modulating IGF-1 and improving antioxidant status. Therefore, the present study aimed to investigate the effects of RT, RSV, and combined RT+RSV on IGF-1, malondialdehyde (MDA), and total antioxidant capacity (TAC) in a murine model of breast cancer. Methods: This experimental study was conducted using a post-test design with a control group. Fifty female Wistar mice were randomly assigned to five groups: healthy control (HC), cancer control (CC), resistance training (RT), resveratrol supplementation (RSV), and combined resistance training plus resveratrol (RT+RSV). The resistance training protocol was performed for 8 weeks, three sessions per week. Resveratrol was administered daily at a dose of 25 mg/kg body weight. At the end of the intervention period, IGF-1 gene expression was assessed using RT-qPCR, while MDA and TAC levels were measured using colorimetric assays. Data were analyzed using two-way analysis of variance (ANOVA). Results: Compared with the HC group, the CC group exhibited significantly increased IGF-1 expression and MDA levels, along with a significant reduction in TAC (p < 0.001). Both RT and RSV interventions independently decreased IGF-1 expression and MDA levels while increasing TAC (p < 0.001), with the greatest improvement observed in the combined RT+RSV group. Significant main effects of resistance training and resveratrol were found for all three variables (p < 0.001), whereas the interaction effect of RT × RSV was significant only for TAC (p < 0.001). Conclusion: The combination of resistance training and resveratrol supplementation may serve as a complementary non-invasive strategy for improving oxidant–antioxidant balance and modulating the IGF-1 axis in breast cancer
